Background
TREM2 is a cell surface receptor of the immunoglobulin superfamily. TREM2 is a type1 transmembrane protein that shuttles to the plasma membrane where it exerts its cell autonomous biological functions. TREM2 undergoes regulated intramembrane proteolysis (RIP). TREM2 is preferentially expressed in microglia and is functionally required for migration, cytokine release, phagocytosis, lipid sensing, ApoE binding, shielding of amyloid plaques, and microglia proliferation in the brian. Most of the functionally investigated mutations are located within the Ig-like domain of TREM2
