BackgroundBTK, also known as Bruton’s Tyrosine Kinase, is a non-receptor tyrosine kinase for B lymphocyte development, differentiation, and signaling. The binding of an antigen to the B-cell antigen receptor (BCR) triggers signaling leading to the activation of the B-cell. Once the BCR is engaged, PLCG2 phosphorylates at several sites, initiating signaling pathway through calcium mobilization, and then activates protein kinase C (PKC) family members. Phosphorylation of PLCG2 performs in tangent with the adapter protein B-cell linker protein BLNK. BTK also works to gather diverse signaling proteins and is implicated in cytokine receptor signaling pathways, as well as function of immune cells of innate as well as adaptive immunity, as part of the Toll-like receptor (TLR) pathway. BTK is involved with signaling pathways that link TLR8/9 to NF-kappa-B. BTK is also needed in order to form functional ARID3A DNA-binding complexes.
